==Example Qiaquick Columns== Taking only column per tube (4 total) with ~8 uL each. This way I can measure the yield. I have two current hypotheses: (I) Previous PCRs have had this much DNA but since we immediately moved to columns, we lost anything greater than 10 ug (high yield) (II) This result is somehow erroneous, and will be corrected for with column purification (low yield) My expected results to confirm each hypothesis are in parentheses. ===Protocol=== 1. Add 45 uL nuclease free water to 5 uL each sample 2. [[Daniel:Protocols/Qiaquick_Column|Qiaquick Column Purification]] 3. Elute with 40 uL EB ===Nanodrop Results=== {| class="wikitable" <hiddentext>generated with [[:de:Wikipedia:Helferlein/VBA-Macro for EXCEL tableconversion]] V1.8</hiddentext> |- style="font-size:12pt" align="center" | width="65" height="15" | Sample | width="65" | ng/uL | width="65" | uL | width="65" | ng |- style="font-size:12pt" align="center" | height="15" | V4S3 | align="center" | 12.1 | align="center" | 40 | align="center" | 484 |- style="font-size:12pt" align="center" | height="15" | V4S3A | align="center" | 11.4 | align="center" | 40 | align="center" | 456 |- style="font-size:12pt" align="center" | height="15" | V6S3 | align="center" | 10.7 | align="center" | 40 | align="center" | 428 |- style="font-size:12pt" align="center" | height="15" | V6S3A | align="center" | 10.6 | align="center" | 40 | align="center" | 424 |} ===Discussion=== This was an unfortunate result. It would suggest hypothesis (II), that there is a lot of contaminant from the PCR and the column washes it away. Will confirm results [[Daniel:Notebook/HiResChrPaint/2013-4-24|tomorrow]].
