Probe Prep Synthesis (Started 3/26/13)
TBU Gel (from EtOH Precipitation 4/18/2013)
1. Preran gel for ~45 minutes at 250V 2. Ran 2 gels w/5 lanes each (out of 12 well gels) 3. 5.5 uL clean TBE, 0.5 uL sample, 6 uL 2X Urea buffer (0.5 ladder also) 4. Precooked samples at 65C for 8 minutes 5. Loaded all 12 uL into each lane 6. Ran gels for 20 minutes at 250 V 7. Stained with 3 uL SYBR gold for 30 minutes 8. Rinsed, let sit for ~20 minutes *this isn't recommended but I didn't have time to image immediately 9. Imaged in gel doc

This gel result is meant to be compared against the result from 4/10/13. Both methods followed the same dye coupling procedure, followed by purification; either column purification (4/10/13) or ethanol precipitation (4/17/13). As of yet I have no comment on which method produced better results.
