PISA Parameter - Update!
- According to Jay’s Science paper, the size of DNA nanoball (Rolony) is 200~400nm.
- 1 pixel size
- 20x, Zoom = 1, 2048 x 2048 --> 284nm
- 63x, Zoom = 1, 2048 x 2048 --> 90.14nm
- 63x, Zoom = 4, 2048 x 2048 --> 22.54nm
- Currently used area_lower, area_upper
- 20x obj. image : 4, 50 (2 x 2 px ~ 7 x 7px --> 568nm x 568 nm ~ 2um x 2um)
- 63x obj. image : 16, 200 (4 x 4 px ~ 14 x 14px --> 360nm x 360 nm ~ 1.26um x 1.26um)
- 63x obj. image : 64, 800 (8 x 8 px ~ 29 x 29px --> 180nm x 180 nm ~ 642nm x 642nm)
- Need to be adjusted!
- Size for Rolony identification --> 300nm ~ 800nm in diameter
- (Image resolution need to be 4k x 4k, so that 1px size of 20x obj. becomes 142nm x 142nm. But for now, keep the resolution 2k x 2k!)
- 20x obj. image : 3 ~ 16 (~2 x ~2 px ~ 4 x 4 px --> ~426nm x ~426nm ~ 1.1um x 1.1um
- 63x obj. image : 9 ~ 81 (3 x 3 px ~ 9 x 9 px --> ~270nm x 270nm ~ 811nm x 811nm
- 63x obj. image : 100 ~ 1296 (10 x 10 px ~ 36 x 36 px --> 225nm x 225nm ~ 811nm x 811nm
Result
- 20x, 63x Zoom=4 images decreased rolony count a little
- But, 63x images increased rolony count a lot…is this real?
