Background
The purpose of this experiment is to isolate individual chromosomes frozen in the metaphase state - where the DNA is condensed and ordered into discrete chromosomes - so that they may be separated out and analyzed. We hope to accomplish this by first culturing cell lines, then after they have grown enough, mixing colcemid, which will freeze the cells in the metaphase state, into the cell culture medium. After harvesting the cells exposed to colcemid, we use various methods to lyse the cells and separate out the DNA from the rest of the cell material.
