MDA with PGP1F, 12th, Dev v9.1

  • Use mineral oil in valve lines
  • I expect...
    • No any leaking of H2O into flow channel
    • Reduce cross-contamination
    • But the response of valves became slower … maybe oil is compressed when pushing?


Experiment

  • MDA reaction for 6 hr.
  • qPCR run for verification
  • Extract just almost 2uL at each tube and run qPCR.


Result

  • Result : Good
  • Looked more even than previous results.
  • Amount of amplicons are detected more along to the right side of chambers
  • Maybe the templates moved more toward right chambers?
  • Would the amplicons are generated more? if the reaction time is longer...
