Microfluidic Devices for Combo-Lock
This page details the microfluidic devices used for the Combo-lock project. I have tried to include specifications whenever possible.
Drop Seq Device
Some specs for the Drop Seq device from the Macosko et al 2013 paper. Below is an image of a single device. A chip contains several dozen of these devices.
As can be seen, 4 lines are used, 3 inlets and an outlet. Two of the inlets are aqueous, the third is oil for droplet formation.
The second thumbnail image shows the drop seq device junctions more closely. The first junction combines the two aqueous solutions; in the original paper this combines a cell solution with beads in a lysis buffer solution. See Combo-Lock protocols for details on which solutions I use. The second junction combines the mixed aqueous phases with the oil phase. This is where droplet formation occurs. Note that post-mixing there are grooves in the device. This facilitates mixing.
