Room Temperature Polymerization Test
The purpose of this test is to determine whether we can run most of the Drop Seq setup at room temperature. I will keep the aqueous phase on ice since normally this will have cells that we want to preserve. However, I will run the oil and device itself at room temperature. The outlet flow will also not run across the hotplate, thereby shortening the overall distance.
For this experiment I used the VA-004/gel loading dye mix from 03-15. This allows better visualization of the aqueous phase and has been shown to gel over time.
Run Parameters
- Line 1: Oil (on ice); X psi; ~2 mL
- Line 2: Aqueous (VA-044P; on ice); X psi; ~1 mL
