PBS/Tween 20 washing[edit]

The following protocol was used to allow water to spread more easily on the microwells

  1. Add 20 uL of Tween 20 to 180 uL H20 (10% dilution)
  2. Add 19.8 mL of PBS to this solution. Mix thoroughly by shaking. The solution should look soapy. This leads to a final dilution of .1% Tween 20.
  3. Wash microwell in solution for 5 min
  4. Remove microwell, dry out on kimwipe
  5. When dry, add silicon gasket and then 30 uL of water to a well to see how it spreads


Results[edit]

The water spread very well over the microwell. 30 uL seems to be enough of a volume to cover the entire square. Some of the water seeped through the silicon gasket. Futhermore, some of the microwell arrays peeled off.


Discussion[edit]

Next time, wait for the microwells to dry more to get a better seal with the gasket. Also, wash in solution for less time (2-3 min) so that the arrays do not peel off.
